A recent study suggests that a naturally occurring protein called Piezo1 may hold promise as a potential treatment for asthma attacks by regulating immune cells within the lungs.
Piezo1 has been identified as a key factor in preventing the overactivation of a specific type of immune cell known as type 2 innate lymphoid cells (ILC2s) in response to allergens. When triggered by allergens, ILC2s initiate an inflammatory cascade in the lungs, leading to airway swelling and constriction, which exacerbates asthma symptoms.
Researchers conducted experiments using an experimental drug called Yoda1, which activates Piezo1, to investigate its impact on ILC2 activity in mice. The results revealed that Yoda1 effectively reduced the activity of ILC2s, thus alleviating asthma symptoms in the mice.
Lead researcher Omid Akbari, a professor at the University of Southern California’s Keck School of Medicine, stressed the significance of targeting ILC2s in allergic asthma and underscored the need for novel mechanism-based approaches to address this inflammation-inducing process in the lungs.
Further investigations unveiled that Piezo1, naturally produced by activated ILC2s, acts to limit their activity. In the absence of Piezo1, ILC2s exhibited heightened responsiveness to allergens, leading to increased lung inflammation.
Of note, similar effects were observed in human ILC2s, suggesting the potential applicability of these findings to human asthma patients. Treatment with Yoda1 demonstrated a reduction in airway hyperreactivity and lung inflammation in mice engineered with human immune cells.
These promising findings indicate that targeting Piezo1 with specific drugs like Yoda1 could offer a therapeutic approach to modulate ILC2 function and alleviate symptoms associated with ILC2-dependent airway inflammation in humans. Future research endeavors will focus on developing targeted drugs to regulate Piezo1 in humans, with the goal of controlling or preventing allergic asthma attacks.
